Human vascular endothelial growth factor receptor (VEGFR) enzyme-linked immunosorbent assay (ELISA) kit instruction manual
The experimental principle of this kit is based on the double-antibody sandwich ELISA method to quantify human vascular endothelial growth factor receptor (VEGFR) in a given sample. The process begins with coating a microtiter plate with a purified monoclonal antibody specific for VEGFR, creating a solid-phase antibody. After blocking non-specific binding sites, the sample containing VEGFR is added to the wells. The VEGFR present in the sample then binds to the immobilized antibody. Next, a horseradish peroxidase (HRP)-labeled secondary antibody is introduced, which recognizes and binds to the captured VEGFR, forming a complete immune complex: antibody-antigen-enzyme-labeled antibody. Following a thorough washing step to remove unbound components, the substrate TMB (3,3',5,5'-tetramethylbenzidine) is added. Under the catalytic action of HRP, TMB turns blue, and the addition of an acidic stop solution changes the color to yellow. The intensity of the yellow color is directly proportional to the concentration of VEGFR in the sample. The optical density (OD) at 450 nm is measured using a microplate reader, and the VEGFR concentration is determined by comparing the OD values to a pre-established standard curve. This method ensures high specificity and sensitivity, making it ideal for accurate quantification of VEGFR levels in biological samples.
