Before using this kit, please read the entire package insert carefully. This ELISA (Enzyme-Linked Immunosorbent Assay) is designed to measure CEACAM1 levels in various biological samples. The principle of the assay involves binding of CEACAM1 to specific antibodies coated on a microplate, followed by detection with an HRP-conjugated secondary antibody. A chromogenic substrate is added, and the reaction is stopped to determine the optical density (OD), which correlates with the concentration of CEACAM1 in the sample.
**Intended Use:**
This kit is intended for research purposes only and not for diagnostic or clinical procedures. It allows for the accurate quantification of human CEACAM1 in biological samples such as serum, plasma, and tissue homogenates.
**Principle of the Assay:**
The standards and samples are incubated in the wells of a pre-coated microplate. After washing, an HRP-conjugated antibody is added, followed by a chromogen solution. The reaction is terminated with a stop solution, and the OD is measured at 450 nm. A standard curve is generated from the known concentrations of the standards, allowing the calculation of unknown sample concentrations.
**Sample Collection and Storage:**
- **Serum:** Collect using a serum separator tube. Allow clotting for 2 hours at room temperature or overnight at 4°C before centrifugation at 2000×g for 20 minutes. Store at -20°C. Avoid repeated freeze-thaw cycles.
- **Plasma:** Use heparin as an anticoagulant. Centrifuge within 30 minutes at 2000×g for 30 minutes at 2–8°C. Store at -20°C.
- **Tissue Homogenate and Other Fluids:** Centrifuge to remove particulates. Assay immediately or store at -20°C. Ensure no hemolysis or contamination occurs.
**Materials Required but Not Supplied:**
- 37°C incubator
- Microplate reader (450 nm)
- Pipettes, disposable tips, and absorbent paper
- Distilled or deionized water

- Standards (6 vials, 0.5 ml/vial)
- Sample Diluent (6.0 ml / 3.0 ml)

**Precautions:**
- Do not mix reagents from different kits.
- Allow all reagents to reach room temperature before use.
- Do not use beyond expiration date.
- Use only deionized or distilled water.
- Keep microplate in its sealed bag until ready for use.
- Wear gloves and lab coat during the procedure.
- Dispose of waste properly after inactivation for 30 minutes.
- Avoid contamination of substrate solutions.
- Chromogen B contains 20% acetone; keep away from heat and flame.
**Reagent Preparation:**
Dilute 1 volume of 20X Wash Solution with 19 volumes of water to make 1X. Store at 2–8°C for up to one month.
**Assay Procedure:**
1. Prepare all reagents and standards. Add 50 µL of standard or sample in duplicate.
2. Add 100 µL of HRP-conjugate reagent to each well (except blank). Incubate for 60 minutes at 37°C.
3. Wash 4 times manually or automatically.
4. Add 50 µL of Chromogen A and B to each well. Incubate for 15 minutes at 37°C.
5. Add 50 µL of Stop Solution. Read OD at 450 nm.
6. Plot standard curve and calculate sample concentrations.

- 2–8°C for frequent use

**Note:** If sample values exceed the highest standard, dilute with Sample Diluent and repeat the assay. Always follow safety protocols when handling biological materials.
