Human macrophage inflammatory protein 1 (MIP-1) ELISA kit use method
**Note:**
1. Before use, remove the kit from the refrigerator and allow it to reach room temperature for 15–30 minutes. If the enzyme label is not resealed after opening, store the strip in a sealed bag to maintain its integrity.
2. The concentrated washing solution may crystallize during storage. When diluting, gently heat it in a water bath until fully dissolved. This will not affect the test results.
3. Use a dedicated pipette for each step and regularly calibrate the accuracy to prevent errors. It is recommended to complete the sample loading within 5 minutes. For large numbers of samples, consider using an automatic pipetting device to ensure consistency and efficiency.
4. Always prepare a standard curve simultaneously with each test. It is advisable to run duplicate wells for better accuracy. If the sample's concentration is too high (OD value exceeds that of the first standard well), perform a preliminary dilution (n times) before testing. When calculating, multiply the final result by both the dilution factor and the original dilution factor (×n×5).
5. The sealing film is for single-use only to prevent cross-contamination between different tests.
6. Keep the substrate away from direct light to maintain its stability and performance.
7. Follow the manual instructions carefully. The final test results must be based on readings from the microplate reader, not visual estimation.
8. All samples, washes, and waste materials should be treated as biohazardous and disposed of according to local regulations.
9. Do not mix components from different batches of the reagent, as this may lead to inaccurate results.
10. In case of discrepancy between the English and other language manuals, the English version takes precedence.
**Storage Conditions & Shelf Life:**
- Store the kit at 2–8°C.
- The shelf life is 6 months from the date of manufacture.
It’s important to handle all reagents with care and follow the protocol strictly to ensure reliable and reproducible results. Always check the expiration date before starting the experiment, and keep the kit in a clean, dry environment when not in use.
